尼尔·麦克唐纳1966年出生在美国马萨诸塞州的泽道切斯，父母都是爱尔兰移民，家中共有五个兄弟姐妹。1988年雪城大学毕业（艺术学士学位）之后曾在英国伦敦西区著名的表演学校LAMDA（London Academy of Dramatic Arts and Sciences）短暂进修过一段时间。此这里起步，尼尔·麦克唐纳出言过多部戏剧，包括《Cheap Talk》、《Foreigner》、《皆大欢喜》、《Rivals》、《仲夏夜之梦》、《秃头歌女》以及《等待老左》(Waiting for Lefty)等等，1991年凭借《Away Alone》获得Best Actor Dramalogue奖。

尼尔从小就是“星际旅行”科幻系列的粉丝，1996年则有幸在第这一系列的第八部影片《星际旅行：第一类接触》中出演了Lieutenant Sean Hawk一角，此外他还开始在多部电视剧中出境，但一直没能有突出的成绩；尼尔曾一度打算放弃演艺事业，直到经纪人替他争取到了为影片《兄弟连》试镜的机会，结果他在里面成功扮演了巴克康普顿上尉的角色，这也让他接下来继续同斯皮尔伯格以及汤姆·克鲁斯合作《少数派报告》一片。2006年，尼尔亮相的三部影片包括阿尔·帕西诺的《88分钟》、美式主旋律影片《惊涛大冒险》以及受到高度赞誉的《父辈的旗帜》，07年则有同后辈林塞·洛翰合作的惊悚片《双面疑杀》（I Know Who Killed Me）、以及章子怡和丹尼斯·奎德主演的《骑士》等影片。
